Where can I find a charmable Black Widow? I do know you can find them in uw but I wonder where they are located exactly in uw as I have never seen them there.

Only in the Underworld. The black widows will spawn when you restore any reapers (the reaper of vale is the easiest probably)

And how do I restore a reaper? Is there a reaper located at the smite run?

You restore Reapers by doing the quests down there.

See here. Black Widows spawns if you finish the quests given by the reapers (summoning the reaper alone will not make the spider spawn), for example this one.
