i suggest doing HM immediately after NM, that way you bang them both out at the same time. Read up about the mission prior to starting build yourself and heros to suit the monster types (e.g. holy damage in sanctum cay for instance). either print a map from wiki, or keep wiki open on the side to follow the map and bonus routes. not to sound elitist, but if forming pug's i would ask each member to ping their build and expect the same of yourself. in this manner, you can at least get a feel for whether or not you might be wasting your time with that particular pug. not to say that a 'bad bar' can't be played well, but its been my experience has been contrary to this supposition. also, i'm not adverse to paying for a run, particulary in the desert as time is money and if someone can do it in 1/2 the time while i'm afk, that's a good deal. before i start a mission, i always send a shout to alliance to see if anyone cares to join. undoubtedly, there is someone who has done it a million times and is willing to lend a hand - just makes things easier (and quicker). keep a texmod up as you go as many of the missions have explorable areas that count toward cartography titles. for that matter, i recommend skill capping while doing the missions which will save you time in the long run.
i've got 9 tyrian missions to complete for legendary guardian title, so if your on pm me and i can help if your doing one of those..

1.- Wiki every zone/mission.
2.- Sab/Discordway.
3.- ???
4.- PROFIT!!!
If you want to be "original", then I don't care.
But on a more serious note, pull well, flag heroes if AoE is expected.
Do vanquishing + cartography at the same time. It really saves a lot of time.
I personally like using Broad Head Arrow as my elite when going H/H.
Know when to retreat. Save DP removals for these tricky situations.

Use a good ranger build with interrupts. Use SabWay or DiscordWay.

Don't Die. If you do die, compensate for your lack of ability with PvE skills.

Get decent builds. Tyria isn't all that hard, so once you/your heroes have worthwhile skillbars, you should roll though it pretty quickly.
EDIT: Don't PuG. People are idiots, seriously.

Tip one: Play with at least one decent human player whenever possible.
More fun and you can talk over why things failed if that's the case.
Wiki is your friend indeed, specially for tips for the most efficient way to finish a mission.
