I don't really play tombs much but I've played some this weekend and here's some stuff that I found pretty helpful.
On the 'standard' Me/N PD bar with Rend:
-Rend Channeling/Veil/HBoon stacks, and try to divert HBoon/Channeling or in case of channeling interrupt it.
-Interrupt stuff that kills you (in most teams this is for a big part fire eles, searing heat, savannah heat, rodgort's, order of apostacy etc.)
-Interrupt stuff that prevents you from killing (Defensive Anthem, Aegis (not that common but i've ran into it several times), Weapon of Warding etc.)
-Interrupt stuff that prevents you from running relics (Water snares, grasping earth, ward against foes, etc.)
-Against pure spike builds like rangerspike use your cry to interrupt spikes, and try and divert spike skills.
Other than that a lot of the times it depends very much on the situation, some teams bring very specific skills that you need to counter and sometimes you need to play defensive or offensive.
For the rest I'd advice you to just play and you'll pick it up soon enough.

Id say 90% of interrupting is down to pings and the rest just knowing the game, human reaction times vary from 0.15s to 0.25s but you will find that people play with pings between 30ms and 1000ms which makes interrupts almost purely ping related.
If you dont live in central europe give up on hitting 3/4 seconds and just settle for the easy rupts.
The only people i know that get pings under 50 are dutch or polish, everyone else in europe seems to get 300~, but thats the price you pay for playing a "free" game.

When mesmering a monk take into consideration that without channeling that monk won't last more than 15 seconds (unless you are teamed against really good monks which is rare). You can either interrupt it (1 sec cast should be trivial) or strip it (inspired ench is optimal, so you can use it yourself to support pd spammage). Keep in mind that the HB monk will have it covered with HB. If you keep channeling off both monks, your frontlines and eles will destroy them.
If you are a power block mesmer, camp the prot monk. Guardian and SoA are 1 secs, which means a guaranteed power block. A power blocked prot monk should result in a team wipe.

So far, aside from the one post, most people have focused on interrupting... which honestly isn't a huge part of being a mesmer. Dont get me wrong, interrupting and what to interrupt is important, but a much larger part of being a mesmer is the other 4/8 skills on your bar. (Assuming a normal Dom Mesmer bar, you have Cry, PDrain, your elite, and rez sig for the other 4).
The Main skills on a dom mesmer are:
Diversion - This skill can be up 1/2 the time, 6/12 seconds. A lot of mesmers give in the to urge to spam this skill. The thing about diversion is that if you put it on a human player, any skill you will divert will likely be in the first second after its applied. That said, try and time it when the target will be using a skill. On spike, right when they stand up, etc..
Shame - Same as diversion, you are most likely to hit a target in the first second of application.
Rend - Useful on Spikes, or taking off Channeling/HB
Shatter Enchantment - Use to aid in spikes. If you can time taking off SB, even better, but at the beginning dont sweat the small stuff.
Other common skills on a Mesmer bar :
Well of Profane - Mainly used for altar maps, or to get traction against a heavy enchantment team, using Taint/Aegis, etc. That said, if you aren't facing these, dont waste your energy. You may want an extra point or 2 in inspiration as well, if you plan on using this a lot.
Inspired Enchantment - You dont see this a lot anymore, but it was mainly used as a Counter to Tainted Flesh.

Be prepared to take a lot of shit.
Your team will rage at you to shut down everything, interrupt everything, blame not killing stuff and being killed on you. Switch your focus based on whether your frontline or your backline is bitching more.

Hmm played the game again and noticed something odd
I usually got around .16-.18 with both eyes open or my right eye but consistently got around .12 with only my left eye open. I'm not really sure what the reasoning for this is but I may be playing ranger with one eye open from now on

he means hes gonna play with 1 eye closed cause he thinks it makes him play better... maybe that shit like blind people have better hearing... he has better reactions... maybe if you close both eyes you can interrupt 1/4 casts

On average, I got around 1.6 second interrupts with being semi-high and drunk...
Pretty sure I can get 1.4-1.5 being sober and sheit.

Listen to what people want to have interrupted.
Know when to play defensive or aggressive.
