No, but it's hard to become part of the core team of such a guild.
It takes time to get valued and you need to be around a lot.
And even then people might prefer members they have known for years over newer members.
I've seen this in my old guild and it's one of the reasons I went back with part of the guild to the SNOW alliance.
At least newer people have a fair chance there to team up with other players who also want to play Hard Mode.
Not that they did not have any chance in my old guild but it takes several months to be really seen and valued.
Before that you are welcome to join if there is a free spot but they won't keep a spot open for you.
The moment I split I got several comments like 'but all the people I like to play with are still in XXXX' and '... but I almost got to play with X,Y and Z'.
Even 'you have killed the activity'.
That's in a guild where 35 members went over in a couple of days and only about 10 core players decided to stay behind in the old guild.
That's the influence of the core group.
The newer players will become part of the core if they stay long enough and prove themselfs. I've seen it happen but we are talking about members who have been in the guild for months.
The rest of the members is valued and accepted but there is not as much trust as there is between core members.
If I want to play an elite area HM I know exactly who will be able to do that without a problem.
And if I have 8 or 12 of those I don't need the players I'm not sure about.

I recommend you spend some time researching PvE guilds to find one that best suits your playing style. The guilds listed in this post would be a great starting point. Personally, I <3 [LaZy]

[LaZy] FTL! everytime i run with a lazy we lose. May just be bad luck though.
Ontopic: I agree with what you are saying about core members. That is exactly how my guild works. It's not something we do on purpose. But all of the officers have known eachother for about 2 years. Also, new members seem to be kind of shy at first. We will try to get new members to do HM anything with us their first day and usually no one answers in guild chat. Getting on vent/ts will always help you become one of those "core" members.

As many others, i'll call a guild i'm in Tried out SNOW, was in SMS alliance, tried many other guilds/alliances, and didt feel anywhere like it is in [GoDT]. Maybe we're not "leet" guild, but surely great PvE/high end guild heck, not just any guild can say that they have a "god" in their roster
otherwise i've heard of SMS, SNOW, GoE(the only guild out of those whoa are mostly mentioned, i can say that is really great - ex alliance mates, from LOD alliance), TAM, ZoS, ... GoDT of course
Probably impossible to say "the best" PvE guild *wants PvE ladder*, lol

However, not every well known guild is the best one to join, there are loads of others that can suit you more. Also, Guru is not the be and end all of Guild Wars. Many non english speaking guilds regually perform feats first, fastest and more than guilds known on Guru. But people never hear about them as they by and large dont post here.
So I dont think there is a *best* per se, just well known. The best Guild is one where you feel happy in and can play in the way in which you want to play..

I was in Dark, and full of rich players it wasn't.
The guild leader had a few million in weapons, next to no liquid cash. There were a few scrubs banging around with a few mill (mostly bought on e-bay as they had no clue how to play) and there was Akh and I.

I wouldnt say any guild is "best" though if you want to be in guilds that does elite areas, have fun and just play, (not talking about wellknown, but from my experience), SMS (had great time in their alliance, though yet they all were americans :'( ), DVDF ofc (hey shan), i would also say agro, not that wellknown but good guys .
There are loads more but i mention only a few today :P pick and choose carefully
Of course we do a lot of "elite" pve too :P
(Grj, please dont turn this to a flame thread, i dont like anyone talking bad about any guilds)

I was in Dark as well ~half year ago, left after a month (no ebay money here though )
Which leader do you mean btw? Dark had more than 1 leader if I know well.
