The second mission is called Zen Daijun and there will be a few primary quests leading you up to it. Once you finish it you will be back in Seitung Harbor where you can talk to the deck hand to be transported to the Kaineng Docks explorable area just outside of The Marketplace.

Yes you have to do To Zen Daijun

and no 'ascention' isnt the same as tyria

Well, not really an ascension like in Tyria. However, Nah Pui Quarter (second city mission) is often compared to ascension.

Completeing the quest "Closer to the Stars" and beating Naphui Quarter "ascends" your Canthan characters and grants them access to FoW/UW.
